ABSTRACT

Diabetic retinopathy(DR) and coronary heart disease(CHD) are both major chronic vascular complications that seriously jeopardize the health of the population and often occur together in clinical practice, it is of great clinical value to actively explore the association between the two in the process of disease development and methods of prevention and treatment of modern medicine and traditional Chinese medicine(TCM). According to TCM, the heart and eyes physiologically communicate with each other by taking Qi, blood and veins as bridges, blood stasis obstructing collaterals is the common TCM etiology of DR and CHD, whose mechanism involves inflammation, oxidative stress and endothelial dysfunction. Promoting blood circulation and removing blood stasis plays an important role in the same treatment for different diseases and prevention and treatment of comorbidities, possibly by inhibiting the expression of interleukin-1β(IL-1β), endothelin-1(ET-1) and hypoxia inducible factor-1α/vascular endothelial growth factor(HIF-1α/VEGF), regulating phosphatidylinositol 3-kinases/protein kinase B/mammalian target of rapamycin(PI3K/Akt/mTOR) pathway, initiating adenosine monophosphate(AMP)-activated protein kinase/silent information regulator 1(AMPK/SIRT1) and nuclear transcription factor erythroid 2-related factor 2/heme oxygenase-1(Nrf2/HO-1) signaling pathways, inhibiting Hippo/Yes-associated protein(Hippo/YAP) signaling pathway, inhibiting mitochondrial permeability transition pore and anti-platelet agglutination for treating DR and CHD, which provides a multi-component, multi-pathway and multi-target selection strategies and ideas for the prevention and treatment of DR and CHD by TCM from a biological perspective. Based on this, subsequent studies should focus on constructing clinically relevant comorbidity models, conducting multicenter prospective studies, and fully utilizing artificial intelligence technology to gain a deeper understanding of the relationship between the two diseases, so as to elucidate the mechanism of promoting blood circulation and removing blood stasis in preventing and treating panvascular diseases.

ABSTRACT

ObjectiveTo explore the differences in the protective effects of five formulas for promoting blood circulation and removing blood stasis on the aortic endothelial cells of New Zealand rabbits with heart blood stasis syndrome. MethodEighty New Zealand rabbits were randomly divided into a normal group (n=10) and an experimental group (n=70). The heart blood stasis syndrome model was induced by starvation combined with a high-fat diet and adrenaline in the rabbits of the experimental group. Subsequently, the model rabbits were randomly divided into a model group, a Xuefu Zhuyutang group (3.55 g·kg-1·d-1), a Taohong Siwutang group (2.66 g·kg-1·d-1), a Danshenyin group (1.962 g·kg-1·d-1), a Huoluo Xiaolingdan group (2.80 g·kg-1·d-1), a Shixiaosan group (0.56 g·kg-1·d-1), and a c-Jun N-terminal kinase (JNK) inhibitor (SP600125, 5 μg·kg-1)group. The normal group and the model group received the same amount of distilled water. The rabbits in five Chinese medicine groups were treated correspondingly by gavage, and those in the SP600125 group were injected with 0.5 mL of SP600125-dimethyl sulfoxide diluent. After the treatment, the aorta was collected, and terminal deoxynucleotidyl transferase dUTP nick end labeling (TUNEL) assay was used to detect the apoptosis of aortic endothelial cells. The enzyme-linked immunosorbent assay (ELISA) was used to detect serum levels of tumor necrosis factor-α (TNF-α) and interleukin-6 (IL-6). Western blot was used to detect the protein expression of JNK, phosphorylated JNK (p-JNK), B-cell lymphoma-2 (Bcl-2), Bcl-2-associated X protein (Bax), cysteinyl aspartate-specific protease-9 (Caspase-9), and cysteinyl aspartate-specific protease-3 (Caspase-3) in aortic tissues. Real-time fluorescence-based quantitative polymerase chain reaction (Real-time PCR) was used to detect the mRNA levels of JNK, Bcl-2, Bax, Caspase-9, and Caspase-3 in aortic tissues. ResultFive formulas could improve the apoptosis of aortic endothelial cells to varying degrees. To be specific, Xuefu Zhuyutang and Taohong Siwutang were optimal in efficacy, followed by Huoluo Xiaolingdan, Shixiaosan, and Danshenyin, and SP600125 was the worst (P<0.05, P<0.01). Five formulas could reduce the content of TNF-α and IL-6 (P<0.05, P<0.01), down-regulate the protein expression levels of JNK, p-JNK, Bax, Caspase-9, and Caspase-3 (P<0.05, P<0.01), decrease the mRNA expression levels of JNK, Bax, Caspase-9, and Caspase-3 (P<0.05, P<0.01), and up-regulate the protein and mRNA expression levels of Bcl-2 (P<0.05, P<0.01). ConclusionFive formulas can all reduce the apoptosis of aortic endothelial cells in New Zealand rabbits with heart blood stasis syndrome with different efficacies. It may be related to the different effects of five formulas on the JNK signaling pathway.

ABSTRACT

OBJECTIVE@#To observe the effects of Taohong Siwu Decoction(, THSWD) on the mesenchymal stem cells(MSCs) migration, homing number and cytokine expression in callus during the early process of fracture healing, and to explore the mechanism of THSWD on accelerationg fracture healing by regulating the homing of MSCs in rats.@*METHODS@#A rat model of right femoral shaft open fracture was established. Thirty-two 5-week-old male Sprague-Dawley rats, weighting 110 to 130 g, were divided into control group, low-dose group, medium-dose group and high-dose group by using random number table. Distilled water was given to the control group, and the other groups were given Taohong Siwu Decoction. The rats were gavaged twice a day for 5 consecutive days after surgery. Bone volume/tissue volume(BV/TV) and bone mineral density(BMD) were observed using micro-computed tomography (micro-CT) at 21 days after surgery. At 5 days post-fracture, peripheral blood MSCs from THSWD treated and untreated rats were cultured in vitro. Subsequently, the migration ability of MSCs was observed by cell migration assay. The number of MSCs homing to the callus at the early stage of fracture (5 d) was detected by Immunohistochemistry (IHC). Protein chip was used to detect the expression of cytokines in callus.@*RESULTS@#Micro-CT results showed that BV/TV was higher in the high-dose group than in the medium-dose group (P=0.032), and higher in the medium-dose group than in the low-dose group(P=0.041), with no difference between the control and low-dose group (P=0.651). In addition, there was no difference in BMD between low-dose group and the model group (P=0.671), and lower in the low-dose group than in the medium-dose group(P=0.018), and the medium-dose group was lower than the high-dose group(P=0.008). Cell migration assay showed that THSWD promotes enhanced the migration ability of peripheral blood MSCs. IHC assay revealed that CD45-, CD90+, CD29+ MSCs significantly increased in bone callus after THSWD intervention compared with the control group. Protein chip showed that THSWD promoted the upregulation of CINC-1(×2.91), CINC-3(×1.59), LIX(×1.5), Thymus Chemokine (×2.55), VEGF (×1.22) and the down-regulation of TIMP-1 (×2.98).@*CONCLUSION@#THSWD, a representative formula of "promoting blood circulation and removing blood stasis", can significantly accelerate fracture healing, and its mechanism may be related to enhancing the migration ability of peripheral blood MSCs and up-regulating CINC-1, CINC-3, LIX, Thymus Chemokine, VEGF and down-regulating TIMP-1 in bone callus, which promotes the peripheral blood MSCs homing in the early stage of fracture.

ABSTRACT

Traditional Chinese medicine for promoting blood circulation and removing blood stasis has been widely used in clinical practice. However, due to the diversity of the composition of traditional Chinese medicine and the complexity of its interaction with human body, it is difficult to apply traditional quality control ingredients to characterize its overall efficacy. Systematic traditional Chinese medicine is an effective method for studying the efficacy of traditional Chinese medicine, embodying the dialectical unity of holism and reductionism. Salviae Miltiorrhizae Radix et Rhizoma is a common traditional Chinese medicine for promoting blood circulation and removing blood stasis. In this study, we constructed a multi-dimensional network of "efficacy-pharmacological efficacy-targets-components" based on systematic traditional Chinese medicine, and discussed the discovery of the efficacy markers of Salviae Miltiorrhizae Radix et Rhizoma for promoting blood circulation and removing blood stasis. Firstly, based on the Chinese medicine efficacy-pharmacology database, the most relevant pharmacological actions(boundary) for promoting blood circulation and removing blood stasis(function) were obtained, and the target sets(structure) of the corresponding pharmacological action were obtained by the DrugBank database. Then, STRING database was used to construct protein-protein interaction network(relationship) of targets related to promoting blood circulation and removing stasis, and key targets(elements) in the network were selected by evaluating topological parameters of targets. Finally, the potential efficacy markers of Salviae Miltiorrhizae Radix et Rhizoma were predicted by molecular docking based on the key targets of promoting blood circulation and removing blood stasis. The results demonstrated that salvianolic acid B, salvianolic acid A, tanshinone Ⅱ_A and tanshinone Ⅰ were the potential markers of Salviae Miltiorrhizae Radix et Rhizoma on promoting blood circulation and removing blood stasis. Salvianolic acid B, salvianolic acid A, tanshinone Ⅱ_A had been reported to have anti-platelet aggregation, anti-thrombotic, cardiovascular protection and some other pharmacological functions. Based on systematic traditional Chinese medicine, we have preliminarily predicted the efficacy markers of Salviae Miltiorrhizae Radix et Rhizoma in promoting blood circulation and removing blood stasis in this study, providing a research method for the discovery of efficacy markers and a reference for the overall quality control of traditional Chinese medicine.

ABSTRACT

To a certain extent, put forward the concept of " component of traditional Chinese medicine (TCM)" simplifies the complexity of multi-component and multi-target of TCM, which provides a possibility for the clarification of the material basis of the efficacy of TCM, and also provides a new direction for promoting the modernization and industrialization of TCM, promots the high quality development of TCM. The correlation between prescription and disease syndrome has made rapid progress, both basic research and clinical application are fruitful. However, the correlation between components and disease syndrome still needs to be further studied. The syndrome of blood stasis is a common syndrome of TCM science, and it is more common in various diseases, especially cardiovascular and cerebrovascular diseases, kidney disease, diabetes and hyperlipoidemia. A large number of studies have shown that some specific components contained in TCM or TCM compound can improve the related indexes of patients or experimental animal model with blood stasis syndrome. It is manifested in reducing blood viscosity, inhibiting platelet activation and adhesion aggregation, changing erythrocyte deformability index, inhibiting thrombosis and so on. Blood stasis is not only the pathogenic factor of many diseases, but also the pathological product of many kinds of diseases, which involves a wide range of diseases. Therefore, this study will study the progress of different components of TCM in the prevention and treatment of blood stasis syndrome, focusing on saponins, flavonoids, organic acids, polysaccharides, alkaloids and other active components in improving hemorheological abnormalities, hypercoagulability, platelet activation and adhesion aggregation, thrombosis. Based on the thought of component-disease syndrome, this paper searches the relevant literature in recent 20 years, classifies and summarizes the achievements of different components in the prevention and treatment of blood stasis syndrome, and hopes to provide some ideas for the further study of the pharmacological action of TCM components, the study of compatibility of TCM components and the research of TCM components.

ABSTRACT

Platelet adhesion is a key process in thrombosis. Anti-platelet adhesion effect of some Chinese medicines for promoting blood circulation and removing blood stasis (PBCRBS) has been reported, but their relative efficacies as a whole and specific targets remained unclear. This paper combined activity screening, drug compatibility analysis, pathway clustering, target prediction, and molecular docking to explore the mechanism of anti-platelet adhesion by PBCRBS Chinese medicine. Screening the activity of anti-platelet adhesion of 58 commercially available PBCRBS Chinese patent medicines showed that about 50.0% significantly inhibit ADP-induced platelet adhesion in vitro, and about 96.6% significantly inhibit thrombin-induced platelet adhesion in vitro. The animal experiment involved was approved by the Animal Ethics Committee of Tianjin International Biomedical Research Institute. Combined with the auxiliary platform for TCM (V2.0) inheritance showed that the compatibility of Danshen-Chuanxiong was used most frequently among the top 20 active proprietary Chinese patent medicines. IPA network analysis revealed that IL-1, APP and CCL2 might be the key targets for anti-platelet adhesion function of Danshen-Chuanxiong against atherosclerosis, neuroinflammation and chemokine signaling pathways as the main mechanisms. Molecular docking analysis confirmed the interaction between one of the active compounds shared by Danshen and Chuanxiong, i.e. chlorogenic acid, with its target CCL2. This study provides TCM theory guidance and experimental support for targeting platelet adhesion in anti-thrombosis therapy by Chinese medicine for promoting blood circulation and removing blood stasis.

ABSTRACT

Traditional Chinese medicine for promoting blood circulation and removing blood stasis is a kind of drug with cold or warm medicinal properties which is commonly used in clinical practice. It is an excellent carrier for studying the nature-effect relationship of traditional Chinese medicine. Therefore, this study will acquire the main active components and targets based on the drug-based research method. The Cytoscape 3. 5. 1 platform was used to construct the protein interaction network, and the Bin GO plug-in was used to perform functional annotation and statistical analysis on the identified functional modules. The results showed that the bitter-liver protein interaction network mainly participates in cell cycle process, lipid catabolic process, blood circulation to exert the effect of promoting blood circulation and removing blood stasis; the cold protein interaction network mainly participates in vasoconstriction through targets such as EDNRA,regulates blood coagulation through targets such as PLAU,and thus exerts the effect of cooling blood and eliminating phlegm; warm protein interaction network mainly participates in the regulation of platelet activation through targets such as P2 RY12,thereby exerting the effect of promoting blood circulation, relieving pain and relieving pain. This study explains the common characteristics of the bitter-liver combination and the specific characteristics of cold or warm medicinal properties from the molecular network level, which provides a new idea for the intrinsic relationship between the medicinal properties and efficacy of traditional Chinese medicine.

ABSTRACT

The herbs for promoting blood circulation and removing blood stasis were used commonly in clinical, and most of them are pungent and bitter. In order to study the nature-effect interrelationship of the herbs for promoting blood circulation and removing blood stasis, the TCMSP platform and Cytoscape 3. 5. 1 platform were used to construct warm-pungent-liver and warm-bitter-liver of the complement and intersection protein interaction network and the target-disease network and the network module was analyzed. As a result,warm-liver target-disease network is associated with diseases such as cancer, hypertension, and depression, which exerts the efficacy of warming Yang and transforming Qi,promoting Qi and activating blood, removing blood stasis and dispersing phlegm. The bitter taste target-disease network is associated with diseases such as myocardial infarction, cancer, inflammation and other diseases, which exerts the efficacy of dissipating the stasis. The pungent taste target-disease network is associated with diseases such as cancer, cardiovascular disease, osteoporosis and other diseases, which exerts the efficacy of invigorating the circulation of blood and eliminating stagnation. The research shows that the medicinal combination of warm-pungent-liver and warm-bitter-liver has the efficacy of promoting blood circulation and removing blood stasis by regulating different targets in different disease processes.

ABSTRACT

Salviae Miltiorrhizae Radix et Rhizoma and Carthami Flos are commonly traditional Chinese medicines with invigorating blood circulation and eliminating blood stasis, but they are different in effects due to differences in five tastes and four properties. In this study,Salviae Miltiorrhizae Radix et Rhizoma and Carthami Flos are selected as research vectors to obtain the active ingredients and targets through TCMD,TCMSP database and Ch EMBL database. The protein interaction information of the target is used to obtain from STRING online database, then imported into Cytoscape software to plot the protein interaction network and perform GO enrichment analysis. The results show that the heart-liver protein interaction network, involving blood circulation and hepatic lipid metabolism, thereby exerting the effect of activating blood circulation. The cold-bitter protein interaction network involves the biological process of vasoconstriction, thereby exerting cooling blood and the efficacy of eliminating phlegm. The warm-pungent protein interaction network involves blood coagulation, lipid metabolism and other biological processes to play the role of phlegm pain. Through analysis,it is found that the relationship between pharmacological efficacy and medicinal properties has a certain degree of specificity, which facilitates the subsequent scientific and systematic study of medicinal properties on the basis of this study.

ABSTRACT

OBJECTIVE: To study the protective effects of Qizhi tongluo capsules and their 9 ingredients decomposed recipes for promoting blood circulation and removing blood stasis (PBCRBS) on cerebral ischemia-reperfusion injury model rats, and to lay a foundation for studying the active components of Qizhi tongluo capsules through decomposed recipes. METHODS: Totally 60 SD rats were randomly divided into sham operation group (0.5% CMC-Na), model group (0.5% CMC-Na), Qizhi tongluo capsules without the ingredients for PBCRBS group [0.389 g/(kg·d)], the ingredients of Qizhi tongluo capsules for PBCRBS group [0.253  g/(kg·d)] and Qizhi tongluo capsules group [0.500 g/(kg·d)], with 12 rats in each group. The rats were given relevant medicine intragastrically, once a day, for consecutive 14 d. 2 h after last medication, except for sham operation group, cerebral ischemia- reperfusion injury model was induced by suture method in each group. The neurological function score was measured according to Bederson score 2 h after ischemia and 24 h after reperfusion. The area of cerebral infarction was calculated by TTC staining. The contents of NO, MDA, LDH and SOD in cerebral tissue of rats were measured by biochemical method. The contents of IL-1β and TNF-α in cerebral tissue of rats were detected by ELISA. RESULTS: Compared with sham operation group, the neurological function score, the infarct area and the contents of NO, MDA, LDH, IL-1β and TNF-α in cerebral tissue were significantly increased in model group (P<0.05). The content of SOD in cerebral tissue were decreased significantly (P<0.05). Compared with model group, the neurological function score of rats was decreased significantly in each administration group (P<0.05); the area of cerebral infarction, the contents of NO, MDA, LDH, IL-1β and TNF-α in cerebral tissue were decreased significantly (P<0.05), while the content of SOD in cerebral tissue was increased significantly (P<0.05). Compared with Qizhi tongluo capsules group, the area of cerebral infarction, the contents of NO, MDA, LDH, IL-1β and TNF-α in cerebral tissue were increased in the ingredients of Qizhi tongluo capsules for PBCRBS group and Qizhi tongluo capsules without the ingredients for PBCRBS group (P<0.05), the content of SOD was decreased significantly (P<0.05). CONCLUSIONS: The formulation of Qizhi tongluo capsules and their 9 ingredients decomposed recipes for PBCRBS (without 9 ingredients for PBCRBS) have protective effects on cerebral ischemia-reperfusion injury in rats. Although the protective effects of decomposed recipes for PBCRBS is not as good as that of the whole prescription, the protective effects of whole formulation were decreased significantly. It shows that the drugs for PBCRBS play an important role in protecting the function of cerebral ischemia reperfusion injury. It is of certain significance to separate the ingredients for PBCRBS to clarify the effect of Qizhi tongluo capsules.

ABSTRACT

To evaluate the promoting blood circulation and removing blood stasis effects of Danshen-Honghua(DH) herb pair with different preparations (alcohol, 50% alcohol and water) on blood rheology and coagulation functions in acute blood stasis rats, and optimize the best preparation method of DH based on principal component analysis(PCA), hierarchical cluster heatmap analysis and multi-attribute comprehensive index methods. Ice water bath and subcutaneous injection of adrenaline were both used to establish the acute blood stasis rat model. Then the blood stasis rats were administrated intragastrically with DH (alcohol, 50% alcohol and water) extracts. The whole blood viscosity(WBV), plasma viscosity(PV), erythrocyte sedimentation rate(ESR) and haematocrit(HCT) were tested to observe the effects of DH herb pair with different preparations and doses on hemorheology of blood stasis rats; the activated partial thromboplastin time(APTT), thrombin time(TT), prothrombin time(PT), and plasma fibrinogen(FIB) were tested to observe the effects of DH herb pair with different preparations on blood coagulation function and platelet aggregation of blood stasis rats. Then PCA, hierarchical cluster heatmap analysis and multi-attribute comprehensive index methods were all used to comprehensively evaluate the total promoting blood circulation and removing blood stasis effects of DH herb pair with different preparations. The hemorheological indexes and coagulation parameters of model group had significant differences with normal blank group. As compared with the model group, the DH herb pair with different preparations at low, middle and high doses could improve the blood hemorheology indexes and coagulation parameters in acute blood stasis rats with dose-effect relation. Based on the PCA, hierarchical cluster heatmap analysis and multi-attribute comprehensive index methods, the high dose group of 50% alcohol extract had the best effect of promoting blood circulation and removing blood stasis. Under the same dose but different preparations, 50% alcohol DH could obviously improve the hemorheology and blood coagulation function in acute blood stasis rats. These results suggested that DH herb pair with different preparations could obviously ameliorate the abnormality of hemorheology and blood coagulation function in acute blood stasis rats, and the optimized preparation of DH herb pair on promoting blood effects was 50% alcohol extract, providing scientific basis for more effective application of the DH herb pair in modern clinic medicine.

ABSTRACT

Objective To observe the effects of central exudative chorioretinopathy(central exudative chorioretinopathy,CEC)treated with blood-activating and stasis-removing Chinese medicines. Methods All 339 cases of central exudative chorioretinopathy with 346 infected eyes were randomly recruited into a treatment group(176 cases with 181 infected eyes)and a control group(163 cases with 165 infected eyes). The treatment group was treated with traditional Chinese medicine by dialectical combination of syndrome and disease, while the control group was treated with conventional treatment. Results The corrected visual acuity was 0.01~1.2 in the treatment group.Of all 181 eyes in 176 patients,76 eyes were cured(41.98%),85 eyes were improved(46.96%),and 20 eyes were ineffective(11.6%),with a total effective rate being 88.94%.BY contrast,in the control group,of all 165 eyes in 163 patients,62 eyes were cured(37.57%),67 eyes were improved(40.60%),and 36 eyes were ineffective(21.83%),with a total effective rate being 78.17%.There was significant difference in the total effective difference between the two groups(U=2.05693,P<0.05).FFA and OCT were reexamined for the patients in the treatment group three menths after the treatment. The results showed that fluorescein leakage and choroidal neovascularization(choroid al neovascularisation,CNV)lesions disappeared in 76 eyes;fluorescein leakage and CNV lesions relieved in 85 Eyes;fluorescein leakage still existed in 12 eyes;and sear formed in 8 eyes. Conclusion Blood-circulating and stasis-removing Chinese medicines have good effects in promoting subhyaloid hemorrhage and exudation, eliminating retinal oedema, and improving eyesight.

ABSTRACT

Objective To explore the regularity of heat shock protein 70(HSP70)expression and its function in renal ischemia/reperfusion injury(IRI),and to reveal whether traditional Chinese medicine, Xuebijing injection(血必净注射液),induces HSP70 synthesis and has a protective effect on IRI.Methods One hundred and eight female Sprague-Dawley(SD)rats,(200?20)g in weight, were used in this experiment, and randomly divided into three groups(each n=36):control(A),model(B)and treatment group(C).The I/R model was established by clamping renal pedicles on both sides for 45 minutes to cause ischemia and then reperfusion was made.In group A,a similar model procedure was performed, but without ischemia. At 5-10 minutes before the IRI was performed,5 ml/kg of normal saline and 5 ml/kg of Xuebijing injection was injected through the femoral vein in group B and C respectively. In B and C groups, according to the durations of reperfusion for 0,6,12,24,36 and 48 hours, the rats were subdivided into six subgroups (each n=6).At the end of each time point, blood and renal tissue were collected to measure blood creatinine (SCr)and urea nitrogen(BUN),and Western blotting method was used to examine the expression of HSP70. In another kidney, renal tissue was obtained for hematoxylin and eosin(HE)staining pathological and immunohistochemical examinations. Results In B and C groups, the SCr and BUN levels at 6 hours after I/R were significantly higher than those in the A group, and those in the C group were lower than those in the B group(P
